USED NASTED-PCR DETECTION OF PHYTOPLASMA CAUSING BIG BUD DISEASE ON TOMATO IN IRAQ

Symptoms similar to those produced by the phytoplasma disease "Big bud" were observed on tomatoes (Solanum lycopersicum L.) grown in field Mosul city, Iraq. Diseased plants characterized twisting, corrugated, yellowing or reddening of leaves. The sepals flowers acquired hypertrophied form, fused together and created a bell-shaped sterile bud (phyllody) green anthocyanin color. stems lignified, phloem necrosis was stem. In mid-September 2020, samples 30 diseased 2 healthy (control) tomato collected from fields. Phytoplasmas detected PCR Nested-PCR 10 samples, use universal specific primers. results phylogenetic tree consisting 27 genetic sequences 16 S ribosomal RNA gene Candidatus Phytoplasma trifolii, locally isolated Iraq/Mosul, showed sequence identity with high percentage similarity 99% trifolii 16Sr member VI-A. Clover proliferation group (16Sr VI), various isolates around world.
